Basic plot: British "serial killer" makes their way to America, pursued by Sherlock. This "serial killer" turns out to be some supernatural creature and the Winchesters show up to the crime scene. Sherlock of course immediately sees through them, and they probably end up in jail. Sherlock is curious as to their motives to impersonate FBI agents, and ends up talking to them and eventually they all end up chasing the creature together. Still trying to figure out how the Doctor fits into the picture.
Marc Reimer Interesting plot twist: Cas decides that its violent and disruptive to normal Earth life, and decides the best thing to do is kill it. He tries to use his angel powers and smite it, but it doesn't work
Marc Reimer So that's how the Doctor comes in!! Word gets to him of this rampaging alien (that no one can kill) and now he has to come and deal with it
Marc Reimer That would be super cool! Maybe each one has a little bit more story tacked onto the end that the last one didn't have to keep things a bit more interesting

Marc Reimer Oh crud. That sounds like it could be bad. If that happened, this special might be expanded into a two-parter.
